This kit recognizes natural and recombinant Human DKK1. No significant cross-reactivity or interference between Human DKK1 and analogues was observed.

Samples: Serum, plasma and other biological fluids. Assay Type: Sandwich. Detection Range: 31.25-2000pg/mL. Sensitivity: The minimum detectable dose of Human DKK1 is 18.75pg/mL (The sensitivity of this assay, or lowest detectable limit (LDL) was defined as the lowest protein concentration that could be differentiated from zero).

Intended Uses: This ELISA kit applies to the in vitro quantitative determination of Human DKK1 concentrations in serum, plasma and other biological fluids.

Principle of the assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to DKK1. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for DKK1 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain DKK1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of DKK1. You can calculate the concentration of DKK1 in the samples by comparing the OD of the samples to the standard curve.

This gene encodes a protein that is a member of the dickkopf family. It is a secreted protein with two cysteine rich regions and is involved in embryonic development through its inhibition of the WNT signaling pathway. Elevated levels of DKK1 in bone marrow plasma and peripheral blood is associated with the presence of osteolytic bone lesions in patients with multiple myeloma. [provided by RefSeq, Jul 2008]

DKK1: Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero- posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease. Interacts with LRP6.
