CH1 reacts strongly with striated muscle forms of tropomyosin, including cardiac alpha-tropomyosin and skeletal alpha- and beta-tropomyosins, but not with tropomyosin isoforms from gizzard (smooth muscle) or chicken embryo fibroblasts. CH1 interferes with the binding of tropomyosin to actin. CH1 recognizes sarcomeric Tm soforms alpha-fast, beta, and alpha-slow with poor affinity for alpha-slow.
